Biotechnological Enhancement of Medicinal Properties in Chrysanthemum sp. Callus Through Salicylic Acid Elicitation

Elicitors are biological or chemical compounds that stimulate plant defense responses and significantly enhance the production of secondary metabolites. Salicylic acid (SA) is one of the important elicitors that activates defense pathways, leading to increased synthesis of bioactive compounds such as phenolics, flavonoids, and antioxidants. Chrysanthemum sp., as an important ornamental and medicinal plant, contains valuable antioxidant compounds. Plant tissue culture provides a suitable platform to study the effects of SA on the production of these compounds. In this study, chrysanthemum seeds were sterilized and cultured on MS medium. After growth, leaf explants were used for callus induction on a medium containing 2 mg/L BAP and 2 mg/L 2,4-D under light conditions. Salicylic acid elicitor was added to the medium at concentrations of 0, 50, 100, and 150 µM. Total phenolic content, flavonoid content, antioxidant capacity, and anthocyanin levels of callus tissues were measured. Results showed that increasing SA concentration significantly enhanced the production of total phenolics, flavonoids, antioxidants, and anthocyanins. The highest amounts were observed at 150 µM SA. These increases indicate the regulatory role of SA in activating metabolic and genetic pathways related to bioactive compound synthesis. The findings suggest that salicylic acid as an elicitor can improve the medicinal quality of chrysanthemum by boosting secondary metabolite production and antioxidant activity. This study provides an effective strategy to enhance bioactive compound production in Chrysanthemum sp tissue culture.
