Bioinformatics analysis of cis-regulatory elements in the promoter of the Geranyl Pyrophosphate:olivetolate geranyltransferase (GOT) gene and its relationship with the regulation of the cannabinoid biosynthesis pathway in cannabis (Cannabis sativa L.)

چکیده :
The cannabis plant (Cannabis sativa L.) has attracted research attention due to its potential to produce valuable secondary metabolites, such as cannabinoids. The gene Geranyl Pyrophosphate:olivetolate geranyltransferase (GOT) encodes a key enzyme in the biosynthesis pathway of these compounds. In this study, the GOT gene was retrieved from the UniProt database and aligned to the cannabis genome using BLASTP (E-value ≤1e−5, coverage ≥80%). A 2000-bp upstream region of the transcription start site (TSS) for each variant was extracted using the BioMart tool in Ensembl Plants and analyzed in the PlantCARE database to identify cis-regulatory elements. In total, 14 regulatory elements were identified and classified into four groups: 1) hormone-responsive elements (ABA, SA, auxin, and gibberellin), 2) abiotic stress-responsive elements (drought, light, and wounding), 3) function-specific elements (seed regulation, defense, and stress), and 4) structural elements (core promoter, MYBHv1 binding, and protein binding). The results revealed that GOT1, dominated by core promoter elements, plays a fundamental role in transcription initiation, whereas the variants GOT2, GOT3.1, and GOT3.2 exhibit more dynamic expression in response to stimuli due to the abundance of promoter/enhancer elements. The co-occurrence of hormone- and abiotic stress-responsive elements confirms the role of these genes in environmental adaptation and growth regulation. The identification of seed-specific elements also suggests their involvement in critical developmental stages. These findings were visualized using TBtools software and represent a key step toward understanding the regulatory mechanisms underlying the biosynthesis of secondary metabolites such as THCV and CBDV, which hold significant potential for medical applications.
