Prediction of the effect of thidiazuron, kinetin and BAP on callus induction, somatic embryogenesis and regeneration of Hyoscyamus niger by Box-Behnken design

چکیده :
Hyoscyamus niger, a notable medicinal plant from the Solanaceae family, serves as a valuable source of the alkaloids hyoscyamine and scopolamine. This research utilized the Box-Behnken design within the response surface methodology to determine and forecast the ideal concentrations of TDZ, BAP, and kinetin for achieving callus formation and regeneration through somatic embryogenesis using leaf and petiole explants. The petiole explant was selected for the percentage of callusing and number of the shoots and the leaf explant was selected for callus fresh weight and percentage of shooting prediction. The callusing percentage varied between 20.00% and 100%, while the fresh weight of callus ranged from 70.08 mg per explant to 1287.08 mg per explant. The somatic embryogenesis percentage spanned from 40.00% to 100%, with the shooting percentage falling between 50.00% and 100%. Additionally, the number of shoots per callus ranged from 1.00 to 8.50. The R2 values were as follows: 86.80% for the callusing percentage, 74.47% for the fresh weight of callus, 79.20% for the somatic embryogenesis percentage, 78.40% for the shooting percentage, and 97.01% for the number of shoots. The best conditions for maximizing the percentage of callusing (100%) were predicted by culturing of the petiole explant on the MS medium containing 0.11 mg/L TDZ and 1.253 mg/L BAP. the highest callus fresh weight (3273.64 mg/explant) predicted by culturing the leaf explant on MS medium containing 0.52 mg/L TDZ. The highest percentage of somatic embryogenesis (100%) predicted by culture of petiole explant on MS medium supplemented with 0.533 mg/L TDZ and 1 mg/L BAP or MS medium containing 1.159 mg/L TDZ and 3 mg/L BAP. The best conditions for maximizing the percentage of shooting (100%) were predicted by culturing of leaf explant on MS medium with 0.18 mg/L TDZ and 2.89 mg/L BAP. it is predicted that the highest number of the shoots obtained by culturing the petiole explant on MS medium with 0.1 mg/L TDZ and 3 mg/L BAP (8.61 shoots per callus).
