Evaluation of the Apoptotic and Cytotoxic Effects of Selected Methanolic and Ethanolic Medicinal Plant Extracts on Cancer Cells

Abstract This study evaluates the cytotoxic effects of plant-derived extracts as potential alternatives to conventional cancer therapies. Methanolic and ethanolic extracts from Catharanthus roseus, Melia azedarach, Berberis integerrima, Mahonia fortuni, and Betula pendula were tested on cell lines of KB, HeLa, G292, U87-MG, A431, and MCF-7 and a normal line of HGF-1 using MTT assay and flow cytometry. The non-liposomal methanolic extract of C. roseus showed the highest cytotoxicity (~96%) against MCF-7 cells. Liposomal M. azedarach extract demonstrated over 80% cell death in cancer cells, primarily inducing apoptosis and causing G0/G1 cell cycle arrest. Extracts from B. integerrima and M. fortuni also promoted apoptosis but exhibited significant toxicity toward normal HGF-1 cells. In contrast, B. pendula extract effectively induced apoptosis in KB cells with minimal toxicity to normal cells. Results suggest that plant extracts, especially in liposomal form, have significant potential to selectively induce apoptosis in cancer cells, with liposomal M. azedarach emerging as a promising candidate for further development in combination cancer therapies. Selectivity and low toxicity toward normal cells are critical for their therapeutic potential.
