مطالعۀ بیوانفورماتیکی ژن های منتخب دخیل در مسیر بیوسنتز فلاونوئیدها در زیرۀ سبز

چکیده :
" مطالعۀ بیوانفورماتیکی ژن های منتخب دخیل در مسیر بیوسنتز فلاونوئیدها در زیرۀ سبز " گیاهان دارویی، به علت وجود ترکیبات مختلفی که در طول واکنشهای متابولیکی، در آنها انباشته میشود، دارای خواص دارویی و ادویهای میباشند. مادۀ موثرۀ این گیاهان حاوی انواع متابولیتهای ثانویه از جمله فلاونوئیدها، با خاصیتهای متنوع از جمله، آنتی اکسیدانی میباشد. در تحقیق حاضرپنج ژن منتخب موثر در مسیر بیوسنتز فلاونوئیدی در گیاه داروئی زیرۀ سبز، که براساس مطالعه NGS بیشترین تغییر بیانی در زیرۀ سبز را داشتند، مورد مطالعات بیوانفورماتیکی قرار گرفتند. درنهایت دو ژن 1196 و 32640 که دارای بیشترین فاصله تکاملی از یکدیگر و از دو خانواده مختلف بودند، جهت شناسایی بیشتر، مورد بررسی قرارگرفتند. ژن 1196 از خانواده GT6، یک گلیکوزیل ترانسفراز در مسیر بیوسنتز ترکیبات فنلی و ژن 32640 از خانواده bHLH، یک فاکتور رونویسی در مسیر بیوسنتز آنتوسیانین میباشد. طبق بررسیهای بیوانفورماتیکی، پروتئین کد شدۀ احتمالی توالی 1196، با شباهت بیش از 85 درصدی با پروتئینی از گیاه هویج، در مرحلۀ پایانی سنتز یک فلاونول، با انتقال یک مولکول گلوکز، آن را گلیکوزیله میکند. در این فرایند دو پروتئین فاکتور رونویسی با این پروتئین در تعامل میباشند. ساختار ثانویه احتمالی این ژن براساس توالی پروتئینی هویج، به دست آمد. کمپلکس MBW ترکیبی از خانوادههای پروتئینهای MYB، bHLH و WD40 میباشد که تنظیم ژنهای رمزگذار آنزیمهایی که به طور خاص در مراحل پایانی مسیر منتهی به بیوسنتز آنتوسیانینها و تاننهای متراکم هستند، را بر عهده دارد. پروتئین کد شدۀ احتمالی توالی 32640، با شباهت بیش از 90 درصدی با پروتئین هویج، یک فاکتور رونویسی است که در این مسیر با 10 پروتئین دیگر از خانوادههای MYB و WD40 در تعامل است. ساختار ثانویه احتمالی این ژن بر اساس توالی پروتئینی هویج به دست آمد. واژگان کلیدی: بیوانفورماتیک، زیرۀ سبز، ساختار ثانویه، فلاونوئیدها. "Bioinformatics study of selected genes involved in the biosynthesis of flavonoids in cumin" Medicinal plants have medicinal and spice properties due to the presence of various compounds that accumulate in them during metabolic reactions. The active substance of these plants contains a variety of secondary metabolites, including flavonoids, with various properties, including antioxidant. In the current research, five selected genes effective in the flavonoid biosynthesis pathway in the cumin medicinal plant, which had the highest expression changes in cumin based on the NGS study, were subjected to bioinformatics studies. Finally, two genes 1196 and 32640, which had the greatest evolutionary distance from each other and from two different families, were investigated for further identification. Gene 1196 from the GT6 family is a glycosyltransferase in the biosynthesis pathway of phenolic compounds and gene 32640 from the bHLH family is a transcription factor in the anthocyanin biosynthesis pathway. According to bioinformatics studies, the probable coded protein of sequence 1196, with more than 85% similarity with a protein from carrot plant, glycosylates it in the final step of the synthesis of a flavonol by transferring a glucose molecule. In this process, two transcription factor proteins interact with this protein. The possible secondary structure of this gene was obtained based on the carrot protein sequence. The MBW complex is a combination of MYB, bHLH and WD40 protein families, which is responsible for the regulation of genes encoding enzymes that are specifically in the final stages of the path leading to the biosynthesis of anthocyanins and dense tannins. The putative encoded protein of sequence 32640, with more than 90% similarity to the carrot protein, is a transcription factor that interacts with 10 other proteins from the MYB and WD40 families in this pathway. The possible secondary structure of this gene was obtained based on the carrot protein sequence. Keywords: bioinformatics, cumin, secondary structure, flavonoids
