The effect of some plant hormones on saffron tissue culture and micro corm production

Abstrac Saffron (Crocus sativus L.) is a triploid and sterile plant that can only be propagated through mother corms. Due to the limitations of natural propagation, the application of tissue culture techniques can provide an effective approach to enhance the multiplication rate of this valuable medicinal plant. In this study, to evaluate the effect of growth regulators on micro corm induction and organogenesis, healthy corms were surface-sterilized and cultured on half-strength MS medium supplemented with 6% sucrose. Hormonal treatments included BAP at concentrations of 0, 2, and 4 mg L⁻¹ and NAA at concentrations of 0 and 1 mg L⁻¹, arranged in a completely randomized design with five replications. The results showed that the treatment containing BAP (4 mg L⁻¹) + NAA (1 mg L⁻¹) produced the highest rate of micro corm formation and sprouting, while the control (hormone-free) treatment exhibited the lowest effect. Furthermore, BAP alone significantly increased the number of shoots, but the combination of BAP and NAA not only enhanced the number of shoots but also significantly improved the diameter and weight of micro corms. Overall, the findings suggest that the combination of BAP (4 mg L⁻¹) and NAA (1 mg L⁻¹) can be considered an effective strategy for in vitro propagation of saffron and the production of vigorous micro corm.
