چکیده :
Gastric cancer is among the most prevalent malignancies in Iran and worldwide. Due to its high mortality rate, metastatic potential, and the severe side effects of chemotherapy, researchers are seeking alternative compounds with reduced toxicity and enhanced efficacy. Stachys schtschegleevii Sosn, a member of the Labiatae family, is an herbaceous to semi-shrubby annual or perennial plant. It possesses notable anti-inflammatory and antimicrobial properties and has been traditionally used to treat conditions such as sinusitis, asthma, rheumatism, colds, urinary tract infections, and dysuria. It is also employed for disinfecting the respiratory, urinary, and female reproductive systems. Furthermore, this plant exhibits strong antioxidant activity, and its effects on apoptosis induction and cell cycle inhibition in various cancer cell lines have been reported. In this study, we investigated the inhibitory effects of the aqueous extract of Stachys schtschegleevii and its biosynthesized magnetic nanoparticles on the proliferation of AGS gastric cancer cells. The cytotoxicity was assessed using the MTT assay. Initially, the concentration and number of AGS cells, along with various concentrations of the plant extract and green-synthesized magnetic nanoparticles (confirmed via physicochemical tests), were determined for treatment. Concentrations of 10, 20, 40, 60, 80, and 100 mg/mL of the extract and nanoparticles were applied to 96-well plates containing 50,000 cells per well for 24, 48, and 72 hours. The results demonstrated that both treatments induced cell death in AGS cells, with the calculated IC₅₀ values indicating a stronger inhibitory effect of the magnetic nanoparticles compared to the plant extract alone. These findings suggest a promising anticancer potential of Stachys schtschegleevii against gastric cancer cells, which is further enhanced at the nanoscale, introducing a viable candidate for future therapeutic studies.
