Evaluation of Genetic Variation in Some Genotypes of Foeniculum vulgare Using ISSR and IRAP Molecular Markers

چکیده :
Fennel (Foeniculum vulgare L.) is one of the oldest and most important medicinal plants belonging to the Apiaceae family, widely used in traditional medicine. The present study was carried out to identify genetic diversity in 20 different genotypes of F. vulgare L. using 5 inter simple sequence repeats (ISSR) molecular markers and 5 Inter-retrotransposon amplified polymorphism(IRAP), collected from different geographical regions of the world.The exotic genotypes were obtained from Leibniz Institute of Plant Genetics and Crop Plant Research (IPK). Native landraces were purchased from Pakan Bazr Com., Isfahan, Iran. Leaf DNA was extracted and purified, and the quality and quantity of the samples were confirmed using gel electrophoresis and spectrophotometry. PCR amplification of DNA fragments with ISSR and IRAP primers was performed, and the products were separated by agarose gel electrophoresis. The results showed that the average number of observed alleles (Na) for both ISSR and IRAP markers was 2. The maximum number of effective alleles in ISSR markers (1.69) was obtained with the UBC808 primer, while the maximum in IRAP markers (1.69) was related to the NIKITA primer. The mean Shannon index was estimated as 0.50 for ISSR and 0.51 for IRAP, indicating that IRAP markers had slightly higher discriminatory power for fennel genotypes. The highest observed heterozygosity (H0) in ISSR (0.40) was related to the UBC808 primer, while in IRAP (0.42) it was obtained with the K001 primer.The polymorphic information content (PIC) for ISSR loci ranged from 0.26 to 0.42 (mean = 0.37), with the highest value related to ISSR2 (0.42). For IRAP loci, PIC values ranged from 0.28 to 0.41 (mean = 0.32), with the maximum related to the SUKKULA primer (0.41). The mean PIC values for ISSR and IRAP markers were 0.37 and 0.32, respectively, confirming the higher efficiency of ISSR. The effective multiplex ratio (EMR) averaged 16 for ISSR and 9.80 for IRAP. The marker index (MI), calculated based on polymorphic bands, averaged 5.89 for ISSR and 3.17 for IRAP, with the maximum MI recorded for ISSR11 and K001 primers, respectively. Cluster analysis grouped the fennel genotypes into seven clusters based on ISSR and IRAP data, which were partially confirmed by principal coordinate analysis (PCoA). Overall, these findings demonstrate the presence of substantial genetic diversity among fennel genotypes worldwide and suggest that both ISSR and IRAP markers are effective tools for studying genetic variability in this species.
