Quantitative and Qualitative Analysis of Tannin Content in Leaf Extracts of a Native Campanula Species from the Eskou Region Using Different Solvents

Campanula species, commonly known as bellflowers, are recognized for their rich phytochemical profile and traditional medicinal applications. This study investigates the total tannin content of leaf extracts from a native Campanula species collected in the Eskou region of East Azerbaijan, Iran. Four solvents—distilled water, 1% hydrochloric acid, 80% ethanol, and 80% methanol—were used for extraction, employing both soaking and boiling methods. Qualitative tests (Ferric Chloride and 10% Sodium Hydroxide) confirmed the presence of tannins, particularly hydrolyzable forms in acidic and alcoholic extracts. Quantitative analysis using the Folin–Ciocalteu method revealed significant variation in tannin yield across solvents, with acidic and methanolic extracts showing the highest concentrations. These findings highlight the influence of solvent type on tannin extraction efficiency and suggest potential applications of Campanula leaf tannins in pharmaceutical and cosmetic formulations.
