Micropropagation of the medicinal plant peppermint

Abstract Mentha piperita L. (peppermint) is an important aromatic and medicinal plant valued for its essential oils, especially menthol and menthone, which are widely used in pharmaceutical, cosmetic, and food industries. Conventional propagation of peppermint through stolons is limited due to low multiplication rates and risk of disease transmission. Tissue culture techniques provide a reliable alternative for the rapid and large-scale production of healthy and genetically uniform plants. In this study, nodal explants of M. piperita were cultured on Murashige and Skoog (MS) medium supplemented with different concentrations of plant growth regulators. Among the tested combinations, MS medium containing 1.0–2.0 mg/L BAP (6-benzylaminopurine) in combination with 0.5 mg/L NAA (α-naphthalene acetic acid) showed the highest shoot induction and multiplication rate. Rooting was achieved on half-strength MS medium supplemented with 0.5 mg/L IBA (indole-3-butyric acid), resulting in a high frequency of root formation. The regenerated plantlets were successfully acclimatized in greenhouse conditions with a survival rate exceeding 85%. The results confirm that in vitro culture of peppermint is an efficient method for mass propagation, germplasm conservation, and future genetic improvement programs.
