شناسایی ژن‌های کلیدی در سلول‌های سرطان پستان تحت تأثیر‎ Withaferin A ‎از گیاه‎ Withania somnifera ‎با ‏بهره‌گیری از تحلیل داده‌های ریزآرایه‌ای

چکیده :

To identify key genes involved in breast cancer cells treated with Withaferin A, relevant ‎microarray data were collected from the Gene Expression Omnibus (GEO) database and ‎analyzed using RStudio software. Gene ontology (GO) and KEGG pathway enrichment analyses ‎were conducted using the ‎DAVID online tool. The protein–protein interaction (PPI) network ‎was constructed using the STRING database, and the differentially expressed genes (DEGs) ‎were imported into Cytoscape software for further analysis. Based on the results, 42 DEGs were ‎identified in the comparison between control group samples and MCF-7 cells, using |log FC| ≥ ‎‎2 as the threshold. According to the KEGG pathway enrichment analysis, mineral absorption ‎‎(hsa04978) was the most significantly enriched pathway. Additionally, based on the gene ‎network reconstruction results using the gene connectivity degree algorithm, four key hub ‎genes (ATF3, DDIT3, IFIT1, and HMOX1) were identified as the most significant hub genes. ‎These hub genes may serve as potential biomarkers for therapeutic targets in breast cancer.‎
