Salinity stress modulates secondary metabolism and antioxidant responses in Dracocephalum moldavica: The synergistic mitigation role of titanium dioxide nanoparticles and arbuscular mycorrhizal fungi

چکیده :
Nowadays, the application of nanoparticles and mycorrhizal fungi has been considered as a strategy to increase plant resistance to environmental stresses. Moldavian balm (Dracocephalum moldavica L.) is an important medicinal plant with significant antioxidant properties. In this study, the effects of salinity, titanium dioxide nanoparticles (TiO₂ NPs), and arbuscular mycorrhizal fungi (AMF) on the defense system of Moldavian balm were investigated using a factorial experiment based on a randomized complete block design. The treatments included salinity at three levels (0, 100, and 150 mM NaCl), TiO₂ nanoparticles at two concentrations (0 and 50 ppm), and mycorrhizal inoculation at two levels (non-inoculated and inoculated with Funneliformis mosseae), with three replications. The results showed that the effects of salinity, TiO₂ NPs application, and AMF on the activity of antioxidant enzymes (ascorbate peroxidase and guaiacol peroxidase) and the concentration of compatible osmolytes (proline and soluble sugars) were significant. The foliar application of TiO₂ NPs and inoculation with AMF increased the activity of antioxidant enzymes at both the 100 and 150 mM salinity levels. The content of soluble sugars and proline increased following the application of 50 ppm TiO₂ NPs, either alone or in combination with F. mosseae, under salinity stress. Moreover, HPLC analysis showed that salinity increased the levels of secondary metabolites, including phenolic compounds (such as p-coumaric acid and caffeic acid) and flavonoids (e.g., quercetin). It appears that the combination of TiO₂ NPs and AMF enhanced stress tolerance in the Moldavian balm plant by stimulating both enzymatic and non-enzymatic antioxidant defense systems.
