In vitro embryo culture in Canna indica L. a method to eliminate long seed dormancy

چکیده :
The search for new medicinal and therapeutic resources is a targeted approach by various researchers in the world's biological sciences to prevent and combat numerous diseases of human society. Canna indica L. is a tropical plant belonging to the Cannaceae family, which is sometimes called Indian shot. Although Canna is more known as an ornamental plant, several reports indicate the medicinal properties of its various parts, which can be promising in programs to introduce new medicinal plants. The seed dormancy period in this plant is very long, and in some reports it is stated to be up to 4 years. For this reason, seed cultivation is justified only for breeding and producing diverse colors of the plant. Developing breeding programs in this plant for various breeding purposes and introducing new varieties and creating genetic diversity requires solutions to reduce germination time and eliminate its long seed dormancy period. In this study, mature and immature seed embryos of Canna (Gol Akhtar – Persian) were sown and evaluated in two cultivation media: full Skoog (MS) and one-half (1/2 MS) in the form of factorial experiments based on a completely randomized design with 5 replications in the summer of 1404. The results indicated the success of the method of planting immature seed embryos in removing the seed dormancy period and producing in vitro seedlings within 25 days at 22°C in a photoperiod of 8 hours of light and 16 hours of darkness compared to fully mature seed embryos. No significant difference was observed between the two cultivation media tested, and the use of a cheaper medium (1/2 MS) is recommended to reduce costs.
