Biotic Elicitation with Yeast Extract Enhances Phytochemical and Antioxidant Profiles of Calendula officinalis Grown In Vitro

Elicitors are effective tools in in vitro culture technology that stimulate plant defense responses, leading to increased production of secondary metabolites. These compounds, whether biotic or abiotic in origin, can activate biochemical pathways and significantly enhance the yield of medicinal compounds. Their use, especially in medicinal plants, represents a novel approach to increase valuable phytochemicals under controlled laboratory conditions. One important medicinal plant in this context is Calendula officinalis (marigold), which is rich in bioactive phytochemicals such as flavonoids, terpenoids, and phenolic compounds. Besides its broad medicinal properties, this plant is widely used in cosmetic, pharmaceutical, and food industries. Due to its high demand and pharmaceutical importance, propagation and metabolite production through in vitro culture have gained considerable attention. In this regard, the application of elicitors like yeast extract can optimize metabolite production in these systems. The objective of this study was to investigate the effect of biotic elicitation using yeast extract at different concentrations on the phytochemical profile, antioxidant activity, and defense-related enzyme activities (catalase and peroxidase) of Calendula officinalis callus cultured in vitro. After sterilization and seed germination, callus induction was performed on a hormone-supplemented medium containing NAA and BAP. Yeast extract at concentrations of 1000, 1200, and 1400 mg/L was applied in a completely randomized design to the calli. Total phenolic content was measured using the Folin–Ciocalteu method at 735 nm and expressed as mg gallic acid per gram fresh weight. Total flavonoid content was determined by aluminum chloride colorimetric assay at 510 nm and reported as mg quercetin equivalents. Antioxidant activity was evaluated by the DPPH assay at 517 nm and expressed as percentage of radical scavenging. Catalase activity was assessed by monitoring hydrogen peroxide decomposition at 240 nm, and peroxidase activity was measured at 470 nm following reaction with guaiacol and H₂O₂. Analysis of variance revealed that yeast extract as a biotic elicitor significantly enhanced total phenolics, total flavonoids, antioxidant activity, and the activities of catalase and peroxidase enzymes compared to the control. Increasing yeast extract concentrations resulted in significant improvements in these parameters, demonstrating the concentration-dependent efficacy of the elicitor in enhancing medicinal compound production and strengthening defense mechanisms in Calendula officinalis callus under in vitro conditions.
