Comparative Analysis of Phenolic Profiles and Biological Activities in Three Lamiaceae Species (Rosmarinus officinalis L., Mentha piperita L. and Ocimum basilicum L.) Using HPLC and HPTLC

چکیده :
The Lamiaceae family includes numerous aromatic herbs with significant ethnomedicinal value, largely due to their rich phenolic and flavonoid composition. In this study, the phenolic profiles and biological properties of Rosmarinus officinalis L., Mentha piperita L., and Ocimum basilicum L. were systematically compared. Botanical authentication was conducted through macroscopic and microscopic analyses and confirmed by herbarium registration. Spectrophotometric assays confirmed the presence of high levels of phenolic and flavonoid compounds. Advanced chromatographic techniques, including High-Performance Liquid Chromatography (HPLC) and High-Performance Thin-Layer Chromatography (HPTLC), were applied to characterize phytochemical markers. Rosmarinic acid was identified as a major constituent in all three species, while menthol and rutin were prominent in M. piperita, and O. basilicum was particularly enriched in eugenol and other phenolic derivatives. These compounds are recognized for their potent antioxidant, antimicrobial, and antifungal properties. Biological activity screening revealed that all three herbs exhibited strong antioxidant potential as demonstrated by free radical scavenging assays. Furthermore, antimicrobial and antifungal evaluations highlighted the effectiveness of R. officinalis L. and O. basilicum L. against bacterial and fungal strains, while M. piperita L. displayed moderate inhibitory activity. Safety evaluations based on heavy metal and pesticide residue analysesconfirmed compliance with international standards. This comparative investigation emphasizes the importance of phytochemical profiling and bioactivity assessment in Lamiaceae plants, supporting their integration into standardized phytopharmaceuticals for antioxidant and antimicrobial applications.
