The effect of some plant hormones on lemon tissue culture and micro propagation

Lippia citriodora (lemon verbena) is a medicinal and aromatic plant from the Verbenaceae family, valued for its essential compounds such as citral and limonene, widely used in pharmaceutical, food, and cosmetic industries. Conventional propagation of this plant through seeds and cuttings is limited due to poor rooting and genetic variability; therefore, tissue culture provides an efficient alternative for mass and uniform propagation. In this study, leaf and nodal explants were surface-sterilized and cultured on MS basal medium. For callus induction, treatments containing 2,4-D (1.0–2.0 mg L⁻¹) combined with BAP (0.5 mg L⁻¹) resulted in vigorous green callus formation. Shoot organogenesis was optimized on MS medium supplemented with BAP (1.0–1.5 mg L⁻¹) plus NAA (0.1 mg L⁻¹), producing the highest number of shoots (an average of 6.2 shoots per explant). For rooting, IBA (0.5–1.0 mg L⁻¹) was the most effective treatment, achieving over 80% rooting frequency. Acclimatized plantlets showed a survival rate of more than 70% after transfer to sterile soil under greenhouse conditions. Overall, the results indicate that the combination of BAP and NAA for shoot induction and IBA for rooting are the most effective treatments for in vitro propagation of Lippia citriodora, providing a reliable strategy for large-scale and uniform production of this valuable medicinal plant.
