Investigating the antioxidant and antibacterial properties of orange blossom (Citrus aurantium) extract

چکیده :
The escalating prevalence of antimicrobial resistance necessitates the exploration of phytochemical alternatives, particularly from Citrus species renowned for their bioactive secondary metabolites. Citrus aurantium L. flowers represent a valuable botanical resource harboring diverse phenolic compounds with significant biotechnological potential for pharmaceutical applications. This investigation elucidated the antimicrobial and antioxidant efficacies of ethanolic orange blossom (C. aurantium flower) extracts through comprehensive in vitro bioassays. Antimicrobial susceptibility was evaluated via well diffusion methodology against five clinically relevant bacterial pathogens: Escherichia coli, Staphylococcus aureus, Listeria monocytogenes, Streptococcus salivarius, and Acinetobacter spp., with minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) determinations established at 12.5 mg/mL and 25 mg/mL, respectively. Inhibition zone diameters ranged from 10 mm (E. coli) to 15 mm (L. monocytogenes and S. salivarius), demonstrating differential species-specific susceptibility patterns. Radical scavenging capacity was quantified through 2,2-diphenyl-1-picrylhydrazyl (DPPH) assay, yielding an IC₅₀ value of 5.8 mg/mL compared to butylated hydroxytoluene (BHT) standard (0.4 mg/mL), while ferric reducing antioxidant power exhibited 40.67 μmol/L equivalency. The observed bioactivities corroborate the phytochemical complexity inherent to C. aurantium floral tissues, attributable to flavonoid glycosides, phenolic acids, and terpenoid constituents that collectively confer antimicrobial and antioxidant properties through multiple mechanistic pathways, including free radical neutralization and membrane integrity disruption. These findings substantiate the biotechnological valorization of C. aurantium flowers as a promising source for developing novel antimicrobial and antioxidant formulations, warranting further investigation into bioactive compound isolation, structural elucidation, and mechanistic studies to advance translational applications in pharmaceutical biotechnology.
