Evaluation of the Effects of Aqueous Extract of Cymbopogon citratus and Its Silver Nanoparticles on Cell Viability Reduction and Apoptosis Induction in the Acute Lymphoblastic Leukemia Cell Line (Molt-4)

چکیده :
Abstract: Acute lymphoblastic leukemia (ALL) is a malignancy of lymphoid lineage blood cells, characterized by the proliferation of numerous lymphocytes involved in the immune system, with higher incidence in children. This study investigates the cytotoxic and apoptotic effects of Cymbopogon citratus (lemongrass) aqueous extract—a plant known for its potent antioxidant properties and traditional medicinal use—on the Molt-4 leukemia cell line. The MTT assay was employed to assess the impact of the extract and its biosynthesized silver nanoparticles, determining IC₅₀ values of 172 and 60 µg/mL at 24 and 48 hours, respectively. To evaluate early and late apoptosis as well as necrosis, Molt-4 cells were treated with ½IC₅₀ for 48 hours and analyzed using acridine orange/ethidium bromide staining to observe cellular morphology. The results demonstrated significant inhibition of cell viability and induction of apoptosis in Molt-4 cells treated with both the extract and its silver nanoparticles. Given the antioxidant nature of Cymbopogon citratus and the oxidative stress associated with conventional ALL therapies, apoptosis induction via antioxidant-rich botanicals may represent a promising therapeutic strategy for this leukemia.
